John Deere 6610 SE esiteltiin vuonna 2000 6010 SE -sarjan suurimpana mallina. Se oli ensimmäisiä kuusisylinterisiä traktoreita tässä sarjassa, ja sen 6,8-litrainen turboahdettu moottori tuotti vaikuttavat 115 hevosvoimaa. Erityisen merkittävää oli moottorin 471 Nm:n vääntömomentti, joka saavutettiin jo 1495 kierroksella.
Traktorissa oli 16+16-vaihteinen vaihteisto hydraulisella suunnanvaihtajalla, joka tarjosi laajan nopeusalueen 2,4-40 km/h. 65 litran minuuttihydrauliikka ja 5260 kilon nostovoima tekivät siitä monipuolisen työkoneen. Merkittävä parannus oli alhainen 73 desibelin melutaso ohjaamossa, mikä oli huomattavasti pienempi kuin 4-sylinterisissä malleissa.
Traktori oli varustettu sähköisesti ohjatulla nostolaitteella ja kitkalukkoisella etuakselilla. 165 litran polttoainesäiliö takasi pitkän toiminta-ajan, ja 4650 kilon työpaino teki siitä vakaan työkoneen. Lisävarusteena saatava ryömintävaihteisto laajensi käyttömahdollisuuksia entisestään. Erityisen käytännöllinen oli 9,9 metrin kääntöympyrä, joka teki traktorista ketterän kokoonsa nähden.
